HEMOSTASIS, THROMBOSIS, AND VASCULAR BIOLOGY Endothelial [Ca]i signaling during transmigration of polymorphonuclear leukocytes

چکیده
Vascular endothelium plays an important role in regulating the transendothelial migration of polymorphonuclear leukocytes (PMNs). In this study, the intracellular calcium ion ([Ca]i) signaling of endothelial cells (ECs) during PMN transmigration was examined at the single-cell level. Human umbilical vein ECs were cultured on a thin layer of collagen gel. The ECs were labeled with fura-2, immersed in formyl-Met-LeuPhe, and subsequently perfused with fresh buffer to establish a gradient of chemoattractant across the EC monolayer. The entire process of PMN rolling on, adhering to, and transmigrating across the EC monolayer was recorded under both phase-contrast and fluorescence optics. The data showed the following: (1) At high concentration (approximately 3 3 106/mL), both PMN suspension and its supernatant stimulated frequent EC [Ca]i elevations across the monolayer; (2) when used at lower concentration (approximately 5 3 105/mL) to avoid the interference of soluble factors, PMN transmigration, but not rolling or adhesion, was accompanied by EC [Ca]i elevation; (3) the latter EC [Ca]i elevation occurred simultaneously in ECs adjacent to the transmigration site, but not in those that were not in direct contact with the transmigrating PMNs; (4) this EC [Ca]i elevation was an initial and required event for PMN transmigration; and (5) PMNs pretreated with 5,5*-dimethyl-1,2-bis(2-aminophenoxy)ethane-N, N, N*, N*-tetraacetic acid transmigrated with the accompanying EC [Ca]i elevation, but they became elongated in the collagen gel. In conclusion, PMNs induce adjacent EC [Ca]i signaling, which apparently mediates the “gating” step for their subsequent transmigration. (Blood. 2000;96:3816-3822)

Polymorphonuclear leukocytes (PMNs) migrate from the blood into areas of inflammation by binding to the endothelial cells of blood vessels via adhesion molecules. Vascular adhesion protein-1 (VAP-1) is one of the molecules mediating leukocyte–endothelial cell interactions. It is also an endothelial cell–surface enzyme (amine oxidase) that produces reactive oxygen species during the catalytic re...
متن کاملSignaling in leukocyte transendothelial migration.
Under a variety of (patho) physiological conditions, leukocytes will leave the bloodstream by crossing the endothelial monolayer that lines the vessels and migrate into the underlying tissues. It is now clear that the process of extravasation involves a range of adhesion molecules on both leukocytes and endothelial cells, as well as extensive intracellular signaling that drives adhesion and che...

OBJECTIVE Although endothelial CD47, a member of the immunoglobulin superfamily, has been implicated in leukocyte diapedesis, its capacity for intracellular signaling and physical localization during this process has not been addressed in detail. This study examined endothelial CD47 spatiotemporal behavior and signaling pathways involved in regulating T-cell transendothelial migration. APPROA...
متن کاملNeutrophil proteinase 3 acts on protease-activated receptor-2 to enhance vascular endothelial cell barrier function.
OBJECTIVE The principle role of the vascular endothelium is to present a semi-impermeable barrier to soluble factors and circulating cells, while still permitting the passage of leukocytes from the bloodstream into the tissue. The process of diapedesis involves the selective disruption of endothelial cell junctions, which could compromise vascular integrity. It is therefore somewhat surprising ...

Endothelial junctions maintain endothelial integrity and vascular homeostasis. They modulate cell trafficking into tissues, mediate cell-cell contact and regulate endothelial survival and apoptosis. Junctional adhesion molecules such as vascular endothelial (VE)–cadherin and CD31/platelet endothelial cell adhesion molecule (PECAM) mediate contact between adjacent endothelial cells and regulate ...
